Loss-of-function variants in NF1 are known to be pathogenic (PMID: 10712197, 23913538). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with clinical features of NF1-related conditions (PMID: 2152033). ClinVar contains an entry for this variant (Variation ID: 449424). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic. -

The Q1126X nonsense variant has been reported previously in association with neurofibromatosis type 1 in the Human Gene Mutation Database as a variant obtained via personal communication (Stenson et al., 2014; vanMinkelen et al., 2014). This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). In summary, we classify this variant as pathogenic. -

The p.Q1126* pathogenic mutation (also known as c.3376C>T), located in coding exon 26 of the NF1 gene, results from a C to T substitution at nucleotide position 3376. This changes the amino acid from a glutamine to a stop codon within coding exon 26. This alteration has been observed in multiple individuals with a personal and/or family history that is consistent with NF1-related disease (Ambry internal data; van Minkelen R et al. Clin Genet, 2014 Apr;85:318-27; Coleman DM et al. Hum Mol Genet, 2022 Feb;31:334-346). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
