Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). This variant has been observed in individual(s) with clinical features of NF1-related conditions (PMID: 31595648). This variant is not present in population databases (ExAC no frequency). This sequence change replaces methionine with leucine at codon 1149 of the NF1 protein (p.Met1149Leu). The methionine residue is moderately conserved and there is a small physicochemical difference between methionine and leucine. -
