The serine at codon 1578 is replaced by cysteine, an amino acid with dissimilar properties. A different alteration at this position, p.S1578F, was reported in a cohort of individuals with a clinical diagnosis of neurofibromatosis (Bendova S et al. J. Mol. Neurosci., 2007;31:273-9). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
