The c.6791_6792insTGA variant (also known as p.Y2264_N2265insD), located in coding exon 45 of the NF1 gene, results from an in-frame TGA insertion at nucleotide positions 6791 to 6792. This results in the insertion of an extra aspartic acid residue between codons 2264 and 2265. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
