This sequence change replaces asparagine, which is neutral and polar, with serine, which is neutral and polar, at codon 2362 of the NF1 protein (p.Asn2362Ser). This variant is present in population databases (rs146296921, gnomAD 0.06%). This variant has not been reported in the literature in individuals affected with NF1-related conditions. ClinVar contains an entry for this variant (Variation ID: 481941). The p.N2362S variant (also known as c.7085A>G), located in coding exon 47 of the NF1 gene, results from an A to G substitution at nucleotide position 7085. The asparagine at codon 2362 is replaced by serine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
