This variant has been observed in individual(s) with clinical features of neurofibromatosis type 1 (NF1) (PMID: 26740943). ClinVar contains an entry for this variant (Variation ID: 872186). This variant is not present in population databases (ExAC no frequency). This sequence change replaces serine with cysteine at codon 2414 of the NF1 protein (p.Ser2414Cys). The serine residue is moderately conserved and there is a moderate physicochemical difference between serine and cysteine. -

The serine at codon 2414 is replaced by cysteine, an amino acid with dissimilar properties. This alteration was detected in an individual with NF1 or clinical suspicion of NF1 (Bianchessi D et al. Mol Genet Genomic Med, 2015 Nov;3:513-25). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
