Neurofibromatosis, type 1 Uncertain:3

In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). ClinVar contains an entry for this variant (Variation ID: 527416). This variant has not been reported in the literature in individuals affected with NF1-related conditions. This variant is present in population databases (rs754511534, gnomAD 0.0009%). This sequence change replaces proline, which is neutral and non-polar, with arginine, which is basic and polar, at codon 2546 of the NF1 protein (p.Pro2546Arg). -

In silico analysis supports that this missense variant has a deleterious effect on protein structure/function; Observed in an individual with kidney cancer (PMID: 29684080); This variant is associated with the following publications: (PMID: 25486365, 29684080) -
Hereditary cancer-predisposing syndrome;CN230736:Cardiovascular phenotype Uncertain:1
The p.P2546R variant (also known as c.7637C>G), located in coding exon 51 of the NF1 gene, results from a C to G substitution at nucleotide position 7637. The proline at codon 2546 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
