This changes the amino acid from a glutamine to a stop codon within coding exon 52. This alteration was detected in 1/500 individuals with Neurofibromatosis Type 1 (NF1) (Fahsold R et al. Am. J. Hum. Genet., 2000 Mar;66:790-818) In a study of individuals with features suggestive of NF1, p.Q2568* was detected in an individual with optic glioma (Ars E et al. J. Med. Genet., 2003 Jun;40:e82). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
