The c.478C>G (p.Q160E) alteration is located in exon 5 (coding exon 5) of the RAD51D gene. This alteration results from a C to G substitution at nucleotide position 478, causing the glutamine (Q) at amino acid position 160 to be replaced by a glutamic acid (E). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.
Breast-ovarian cancer, familial, susceptibility to, 4 Uncertain:1
This sequence change replaces glutamine with glutamic acid at codon 160 of the RAD51D protein (p.Gln160Glu). The glutamine residue is moderately conserved and there is a small physicochemical difference between glutamine and glutamic acid. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with a RAD51D-related disease. ClinVar contains an entry for this variant (Variation ID: 472611). Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). In summary, this variant has uncertain impact on RAD51D function. The available evidence is currently insufficient to determine its role in disease.
