The glutamic acid at codon 14 is replaced by lysine, an amino acid with similar properties. In one study, this variant was detected in 1/91 Irish breast cancer patients and 0/77 ethnically matched controls (McVeigh ÚM et al. Ir J Med Sci, 2020 Aug;189:849-864). Another study identified this variant in 0/1822 breast and/or ovarian cancer cases and 1/2120 controls from the United Kingdom (Loveday C et al. Nat. Genet., 2011 Aug;43:879-882).This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
