This protein forms distinct complexes with cadherins and desmosomal cadherins and is a member of the catenin family since it contains a distinct repeating amino acid motif called the armadillo repeat. The Asp136His variant in the JUP gene has not been reported as a disease-causing mutation or as a benign polymorphism to our knowledge. Asp136His results in a non-conservative amino acid substitution of a negatively charged Aspartic acid to a positively charged Histidine at a position that is conserved across species. In silico analysis predicts Asp136His is probably damaging to the protein structure/function. Furthermore, the Asp136His variant was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The variant is found in ARRHYTHMIA panel(s). -
