This missense variant replaces glycine with glutamic acid at codon 834 of the BRCA1 protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in individuals affected with breast and/or ovarian cancer (PMID: 26306726, 29470806, 32438681, 33948387). This variant has been identified in 2/251028 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

The p.G834E variant (also known as c.2501G>A), located in coding exon 9 of the BRCA1 gene, results from a G to A substitution at nucleotide position 2501. The glycine at codon 834 is replaced by glutamic acid, an amino acid with similar properties. This alteration has been reported as a variant of unknown significance in breast and ovarian cancer cohorts from a variety of ethnic populations (Minucci A et al. Expert Rev. Mol. Diagn., 2015 Aug;15:1383-403; Singh J et al. Breast Cancer Res Treat, 2018 Jul;170:189-196; Santonocito C et al. Cancers (Basel), 2020 May;12). This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive.
