The p.S387T variant (also known as c.1159T>A), located in coding exon 9 of the BRCA1 gene, results from a T to A substitution at nucleotide position 1159. The serine at codon 387 is replaced by threonine, an amino acid with similar properties. In one study of 83 families with suspected hereditary breast and/or ovarian cancer syndrome, this variant was reported in two relatives with ovarian cancer from an ovarian cancer only family (Muller D et al. Fam. Cancer, 2004;3:15-20). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
This missense variant replaces serine with threonine at codon 387 of the BRCA1 protein. Computational prediction suggests that this variant may have deleterious impact on protein structure and function (internally defined REVEL score threshold >= 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in a family affected with ovarian cancer (PMID: 15131401). However, a multifactorial analysis reported a likelihood ratio for pathogenicity based on segregation of 0.0003 (PMID: 31131967). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
