The p.D330N variant (also known as c.988G>A), located in coding exon 9 of the BRCA1 gene, results from a G to A substitution at nucleotide position 988. The aspartic acid at codon 330 is replaced by asparagine, an amino acid with highly similar properties. This alteration was identified in one individual from a cohort of ovarian cancer patients (Akbari MR, J. Med. Genet. 2011 Nov; 48(11):783-6). This variant was also detected in 1/54 female Chinese patients diagnosed with breast cancer (Shen M et al. Transl Cancer Res, 2019 Apr;8:483-490). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -

This missense change has been observed in individual(s) with breast cancer and ovarian cancer (PMID: 14973102, 21965345, 34326862). ClinVar contains an entry for this variant (Variation ID: 37713). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt BRCA1 protein function with a negative predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
