BRCA1 (HGNC:1100): (BRCA1 DNA repair associated) This gene encodes a 190 kD nuclear phosphoprotein that plays a role in maintaining genomic stability, and it also acts as a tumor suppressor. The BRCA1 gene contains 22 exons spanning about 110 kb of DNA. The encoded protein combines with other tumor suppressors, DNA damage sensors, and signal transducers to form a large multi-subunit protein complex known as the BRCA1-associated genome surveillance complex (BASC). This gene product associates with RNA polymerase II, and through the C-terminal domain, also interacts with histone deacetylase complexes. This protein thus plays a role in transcription, DNA repair of double-stranded breaks, and recombination. Mutations in this gene are responsible for approximately 40% of inherited breast cancers and more than 80% of inherited breast and ovarian cancers. Alternative splicing plays a role in modulating the subcellular localization and physiological function of this gene. Many alternatively spliced transcript variants, some of which are disease-associated mutations, have been described for this gene, but the full-length natures of only some of these variants has been described. The BRCA1 c.341C>G; p.Ser114Cys variant (rs786202620) is reported in the literature in an individual with a personal and family history of breast cancer (Solano 2016). This variant is reported in ClinVar (Variation ID: 185998), and is absent from general population databases (Exome Variant Server, Genome Aggregation Database), indicating it is not a common polymorphism. The serine at codon 114 is moderately conserved, and computational analyses (SIFT, PolyPhen-2) predict that this variant is deleterious. Additionally, another variant at this codon (c.340T>C; p.Ser114Pro) has been reported in an individuals affected with breast cancer (Hedau 2004). However, given the lack of clinical and functional data, the significance of the p.Ser114Cys variant is uncertain at this time. References: Hedau Set al. The p.S114C variant (also known as c.341C>G), located in coding exon 5 of the BRCA1 gene, results from a C to G substitution at nucleotide position 341. The serine at codon 114 is replaced by cysteine, an amino acid with dissimilar properties. This alteration has been reported in an Argentinian patient with breast cancer diagnosed at age 45 with a family history of breast and colon cancer (Solano AR et al. Oncotarget, 2017 Sep;8:60487-60495). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
