The arginine at codon 212 is replaced by tryptophan, an amino acid with dissimilar properties. In one study, this alteration was identified in one unaffected control and in a individual with frontotemporal dementia (FTD) (Yu CE et al. Arch. Neurol., 2010 Feb;67:161-70). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear. - |
