The acetylcholine receptor changes subunit composition shortly after birth when the epsilon subunit replaces the gamma subunit seen in embryonic receptors. This sequence change replaces proline, which is neutral and non-polar, with alanine, which is neutral and non-polar, at codon 295 of the CHRNE protein (p.Pro295Ala). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with CHRNE-related conditions.
