Osteogenesis imperfecta type I Pathogenic:2
- -
This sequence change creates a premature translational stop signal (p.Gly154Trpfs*15) in the COL1A1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in COL1A1 are known to be pathogenic (PMID: 7942841, 9295084, 9443882). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with osteogenesis imperfecta (PMID: 26177859). ClinVar contains an entry for this variant (Variation ID: 425633). For these reasons, this variant has been classified as Pathogenic. -
not provided Pathogenic:1
The c.458dupC pathogenic variant in the COL1A1 gene has been reported in association with osteogenesis imperfecta (Lindahl et al. 2015). This variant causes a frameshift starting with codon Glycine 154, changes this amino acid to a a Tryptophan residue and creates a premature Stop codon at position 15 of the new reading frame, denoted p.G154WfsX15. This pathogenic variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. -
