Hereditary cancer-predisposing syndrome Uncertain:2
The c.-1G>T variant is located in the 5' untranslated region (5’ UTR) of the RAD51C gene. This variant results from a G to T substitution 1 bases upstream from the first translated codon. Based on nucleotide sequence alignment, this position is l / highly conserved in available vertebrate species. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
This variant is located one nucleotide upstream of the ATG start codon for the RAD51C gene, altering a position that is not conserved within the Kozak consensus sequence that functions as the protein translation initiation site. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with hereditary cancer in the literature. This variant has been identified in 2/251276 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
not provided Uncertain:1
Located in a regulatory region; in the absence of functional studies, the actual effect of this sequence change is unknown; Nucleotide is not conserved across species and the substitution has no predicted effect on splicing; Not observed at significant frequency in large population cohorts (gnomAD); Has not been previously published as pathogenic or benign to our knowledge -
