This variant is found in 1/268345 alleles at a frequency of 0.000372% in the gnomAD v2.1.1 database, non-cancer dataset (PM2_supporting). The SpliceAI algorithm predicts no significant impact on splicing. The REVEL meta-predictor score for this variant (0.162) suggests that it does not affect the protein function according Pejaver 2022 thresholds (PMID: 36413997) (BP4). The variant has been identified in the ClinVar** database (3x, as uncertain significance) but it is not present in the LOVD database. Based on currently available information, the variant c.59C>T should be considered as of unknown significance. -

The p.S20F variant (also known as c.59C>T), located in coding exon 1 of the RAD51C gene, results from a C to T substitution at nucleotide position 59. The serine at codon 20 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species.
