This missense variant replaces phenylalanine with serine at codon 99 of the RAD51C protein. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with RAD51C-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
The c.296_297delTCinsCT variant (also known as p.F99S), located in coding exon 2 of the RAD51C gene, results from an in-frame deletion of TC and insertion of CT at nucleotide positions 296 to 297. This results in the substitution of the phenylalanine residue for a serine residue at codon 99, an amino acid with highly dissimilar properties. This amino acid position is well conserved in available vertebrate species; however, serine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be neutral by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Based on the available evidence, the clinical significance of this variant remains unclear. -

This sequence change replaces phenylalanine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 99 of the RAD51C protein (p.Phe99Ser). Information on the frequency of this variant in the gnomAD database is not available, as this variant may be reported differently in the database. This variant has not been reported in the literature in individuals affected with RAD51C-related conditions. ClinVar contains an entry for this variant (Variation ID: 484734). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated.
