The c.556_558dupAAA variant (also known as p.K186dup), located in coding exon 3 of the RAD51C gene, results from an in-frame duplication of AAA at nucleotide positions 556 to 558. This results in the duplication of a lysine residue at codon 186. This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Based on the available evidence, the clinical significance of this variant remains unclear. -
