This variant is denoted RAD51C c.956G>C at the cDNA level, p.Arg319Pro (R319P) at the protein level, and results in the change of an Arginine to a Proline (CGA>CCA). This variant has not, to our knowledge, been published in the literature as pathogenic or benign. RAD51C Arg319Pro was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. Since Arginine and Proline differ in polarity, charge, size or other properties, this is considered a non-conservative amino acid substitution. RAD51C Arg319Pro occurs at a position that is poorly conserved across species, although it is expressed through monotremes, and is not located in a known functional domain. In silico analyses are inconsistent regarding the effect this variant may have on protein structure and function. Based on currently available information, it is unclear whether RAD51C Arg319Pro is pathogenic or benign. We consider it to be a variant of uncertain significance. -

The p.R319P variant (also known as c.956G>C), located in coding exon 7 of the RAD51C gene, results from a G to C substitution at nucleotide position 956. The arginine at codon 319 is replaced by proline, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
