The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.2623G>C has been reported in the literature in one individual affected with breast cancer and/or ovarian cancer (Schubert_2019). The report does not provide unequivocal conclusions about association of the variant with Breast Cancer. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Two ClinVar submitters (evaluation after 2014) cite the variant as uncertain significance. This alteration is reported in an individual affected with ovarian and peritoneal mesothelioma cancer (Schubert S et al. Int J Cancer, 2019 Jun;144:2683-2694). The glutamic acid at codon 875 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
