This variant causes a G to A nucleotide substitution at the +5 position of intron 17 of the BRIP1 gene. Splice site prediction tools predict that this variant may have a significant impact on RNA splicing, although this prediction has not been confirmed in published RNA studies. This variant has been reported in an individual affected with colorectal cancer (PMID: 28135145). This variant has been identified in 3/280396 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
The c.2492+5G>A intronic variant results from a G to A substitution 5 nucleotides after coding exon 16 in the BRIP1 gene. This alteration has been seen in a patient with colorectal cancer (Yurgelun MB et al. J. Clin. Oncol. 2017 Apr;35(10):1086-1095). This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site and may result in the creation or strengthening of a novel splice donor site. RNA studies have demonstrated that this alteration results in an incomplete splice defect; the clinical impact of this abnormal splicing is unknown at this time (Ambry internal data). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
