The p.A776D variant (also known as c.2327C>A), located in coding exon 15 of the BRIP1 gene, results from a C to A substitution at nucleotide position 2327. The alanine at codon 776 is replaced by aspartic acid, an amino acid with dissimilar properties. This variant was identified in a patient diagnosed with high grade serous carcinoma (Andrikopoulou A et al. Front Oncol, 2022 Dec;12:1030786). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
