Genes affected
BRIP1 (HGNC:20473): (BRCA1 interacting helicase 1) The protein encoded by this gene is a member of the RecQ DEAH helicase family and interacts with the BRCT repeats of breast cancer, type 1 (BRCA1). The bound complex is important in the normal double-strand break repair function of breast cancer, type 1 (BRCA1). This gene may be a target of germline cancer-inducing mutations. Uncertain significance, criteria provided, single submitterclinical testingAmbry GeneticsJun 13, 2023The p.R707H variant (also known as c.2120G>A), located in coding exon 14 of the BRIP1 gene, results from a G to A substitution at nucleotide position 2120. The arginine at codon 707 is replaced by histidine, an amino acid with highly similar properties. This alteration has been reported in a cohort of 488 patients with stages I to III breast cancer who were tested with a 25-gene panel test (Tung N et al. J Clin Oncol, 2016 May;34:1460-8). This alteration has been reported with a carrier frequency of 0.0003 in 7636 unselected prostate cancer patients and was not observed in 12366 male controls of Japanese ancestry (Momozawa Y et al. J Natl Cancer Inst, 2020 Apr;112:369-376). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -

Uncertain significance, criteria provided, single submitterclinical testingLabcorp Genetics (formerly Invitae), LabcorpAug 16, 2023In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt BRIP1 protein function. ClinVar contains an entry for this variant (Variation ID: 422038). This missense change has been observed in individual(s) with breast cancer (PMID: 26976419). This variant is present in population databases (rs200313471, gnomAD 0.007%). This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 707 of the BRIP1 protein (p.Arg707His). -

Uncertain significance, no assertion criteria providedclinical testingPreventionGenetics, part of Exact SciencesFeb 12, 2024The BRIP1 c.2120G>A variant is predicted to result in the amino acid substitution p.Arg707His. This variant was reported in an individual with breast cancer (Tung et al 2016. PubMed ID: 26976419) and in an individual with prostate cancer (Momozawa et al 2020. PubMed ID: 31214711, Supplementary Table 6). This variant is reported in 0.0029% of alleles in individuals of Latino descent in gnomAD. This variant has an interpretation of uncertain significance in ClinVar (https://www.ncbi.nlm.nih.gov/clinvar/variation/422038/). At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
