The p.T686A variant (also known as c.2056A>G), located in coding exon 13 of the BRIP1 gene, results from an A to G substitution at nucleotide position 2056. The threonine at codon 686 is replaced by alanine, an amino acid with similar properties. This alteration has been identified in an individual with breast cancer and an individual with prostate cancer (Easton DF et al. J. Med. Genet. 2016 05;53(5):298-309; Ray AM et al. Br. J. Cancer 2009 Dec;101(12):2043-7). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
This missense variant replaces threonine with alanine at codon 686 of the BRIP1 protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in individuals affected with breast cancer and prostate cancer (PMID: 19935797, 26921362). Three of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 251360 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.2056A>G has been reported in the literature in sequencing studies of individuals affected with prostate and breast cancer (example, Ray_2009, Easton_2016). These report(s) do not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome or Fanconi Anaemia Complementation group J.
