Familial cancer of breast Uncertain:2
This variant is classified as a variant of uncertain significance as there is insufficient evidence to determine its impact on protein function and/or cancer risk. -
- -
Hereditary cancer-predisposing syndrome Uncertain:1Benign:1
This alteration is classified as likely benign based on a combination of the following: seen in unaffected individuals, population frequency, intact protein function, lack of segregation with disease, co-occurrence, RNA analysis, in silico models, amino acid conservation, lack of disease association in case-control studies, and/or the mechanism of disease or impacted region is inconsistent with a known cause of pathogenicity. -
This missense variant replaces isoleucine with valine at codon 562 of the BRIP1 protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in individuals affected with ovarian cancer (PMID: 26315354), breast cancer (PMID: 17033622), colorectal cancer (PMID: 28135145) and suspected Lynch syndrome (PMID: 25980754). This variant has been identified in 4/282800 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
not specified Uncertain:1
Variant summary: BRIP1 c.1684A>G (p.Ile562Val) results in a conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 1.2e-05 in 251396 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1684A>G has been reported in the literature in individuals affected with breast cancer, ovarian cancer, or colorectal cancer, without strong evidence for causality (e.g. Ramus_2015, Seal_2006, Yurgelun_2017). These reports do not provide unequivocal conclusions about association of the variant with Breast Cancer. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. c.1684A>G was also detected in one individual from the FLOSSIES database (women aged 70+ without cancer) at a frequency of 0.000101. The following publications have been ascertained in the context of this evaluation (PMID: 26315354, 17033622, 28135145). ClinVar contains an entry for this variant (Variation ID: 182352). Based on the evidence outlined above, the variant was classified as uncertain significance. -
Familial cancer of breast;C1836860:Fanconi anemia complementation group J Uncertain:1
This sequence change replaces isoleucine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 562 of the BRIP1 protein (p.Ile562Val). This variant is present in population databases (rs45533636, gnomAD 0.003%). This missense change has been observed in individual(s) with breast and/or ovarian cancer (PMID: 17033622, 25980754, 26315354, 28135145). ClinVar contains an entry for this variant (Variation ID: 182352). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt BRIP1 protein function with a negative predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

Not observed at a significant frequency in large population cohorts (Lek 2016); In silico analysis, which includes protein predictors and evolutionary conservation, supports that this variant does not alter protein structure/function; Observed in individuals with breast, colorectal, and Lynch syndrome-associated cancer and/or polyps, but also in controls (Seal 2006, Yurgelun 2015, Easton 2016, Ramus 2015, Yurgelun 2017); This variant is associated with the following publications: (PMID: 26921362, 25980754, 28135145, 17033622, 26315354) -
