The glutamine at codon 554 is replaced by glutamic acid, an amino acid with highly similar properties. This alteration was reported in two HBOC families; one of these families reportedly had co-occurrence with a likely pathogenic variant in BRCA1 (Li J et al. J. Med. Genet.2016 Jan;53(1):34-42). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |

Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 8e-06 in 251322 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1660C>G has been reported in the literature in individuals/families affected with Hereditary Breast And Ovarian Cancer Syndrome (e.g. Easton_2016, Li_2016, Dorling_2021) but it was also reported in unaffected controls (Dorling_2021). In one affected family, a co-occurring pathogenic variant was detected (BRCA1 c.547+1G>T; Li_2016). These reports do not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Four ClinVar submitters (evaluation after 2014) cite the variant as uncertain significance.
