This missense variant replaces alanine with threonine at codon 453 of the BRIP1 protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. In a large breast cancer case-control study, this variant was absent in 60466 individuals affected with breast cancer and detected in 1/53460 controls (OR=0.442, 95%CI 0.015 to 13.178; PMID: 33471991). This variant has been identified in 5/247722 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
The p.A453T variant (also known as c.1357G>A), located in coding exon 9 of the BRIP1 gene, results from a G to A substitution at nucleotide position 1357. The alanine at codon 453 is replaced by threonine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
