Hereditary cancer-predisposing syndrome Uncertain:2
Uncertain significance, criteria provided, single submitter | clinical testing | Color Diagnostics, LLC DBA Color Health | May 12, 2023 | This missense variant replaces asparagine with serine at codon 370 of the BRIP1 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in individuals affected with ovarian cancer (PMID: 24448499, 26689913) or head and neck squamous cell carcinoma (PMID: 28678401). This variant has been identified in 2/251326 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. - |
Uncertain significance, criteria provided, single submitter | clinical testing | Ambry Genetics | Sep 06, 2022 | The p.N370S variant (also known as c.1109A>G), located in coding exon 7 of the BRIP1 gene, results from an A to G substitution at nucleotide position 1109. The asparagine at codon 370 is replaced by serine, an amino acid with highly similar properties. This alteration has been reported as a missense variant in both germline and tumor tissue of 1 individual from a cohort of 387 ovarian cancer cases (Kanchi KL et al. Nat Commun 2014; 5:3156). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
Familial cancer of breast;C1836860:Fanconi anemia complementation group J Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Jan 08, 2024 | This sequence change replaces asparagine, which is neutral and polar, with serine, which is neutral and polar, at codon 370 of the BRIP1 protein (p.Asn370Ser). This variant is present in population databases (rs777511615, gnomAD 0.0009%). This missense change has been observed in individual(s) with ovarian cancer and an individual affected with head and neck cancer (PMID: 24448499, 28678401). ClinVar contains an entry for this variant (Variation ID: 483144). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt BRIP1 protein function with a positive predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. - |

Fanconi anemia complementation group J Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Illumina Laboratory Services, Illumina | Apr 27, 2017 | This variant was observed as part of a predisposition screen in an ostensibly healthy population. A literature search was performed for the gene, cDNA change, and amino acid change (where applicable). No publications were found based on this search. Allele frequency data from public databases did not allow this variant to be ruled in or out of causing disease. Therefore, this variant is classified as a variant of unknown significance. - |
not provided Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Women's Health and Genetics/Laboratory Corporation of America, LabCorp | Jun 05, 2017 | Variant summary: The BRIP1 c.1109A>G (p.Asn370Ser) variant located in the P-loop containing nucleoside triphosphate hydrolase domain (via InterPro) involves the alteration of a conserved nucleotide and 2/4 in silico tools (SNPsandGO not captured due to low reliability index) predict a benign outcome. However, these predictions have yet to be functionally assessed. This variant was found in 1/122506 control chromosomes at a frequency of 0.0000082, which does not exceed the estimated maximal expected allele frequency of a pathogenic BRIP1 variant (0.0000625). Multiple publications have cited the variant in affected cohorts, however, limited information is available (ie, lack of co-occurrence and cosegregation data). The variant of interest has not, to our knowledge, been reported by reputable databases/clinical diagnostic laboratories. Therefore, until additional information becomes available, ie, clinical and functional studies, the variant of interest has been classified as a "Variant of Uncertain Significance (VUS)." - |
Familial cancer of breast Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Myriad Genetics, Inc. | Feb 27, 2023 | This variant is classified as a variant of uncertain significance as there is insufficient evidence to determine its impact on protein function and/or cancer risk. - |
