Uncertain significance, criteria provided, single submitter | clinical testing | Color Diagnostics, LLC DBA Color Health | Nov 22, 2022 | This missense variant replaces glutamine with glutamic acid at codon 227 of the BRIP1 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in several individuals affected with breast cancer, as well as in an unaffected control individual (PMID: 26921362, 31822495). In a large breast cancer case-control study, this variant has been observed in 5/60466 cases and 5/53461 unaffected controls (OR=0.884, 95%CI 0.256 to 3.054) (PMID: 33471991; Leiden Open Variation Database DB-ID BRIP1_000027). This variant has been identified in 9/250948 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. - |

Uncertain significance, criteria provided, single submitter | clinical testing | Ambry Genetics | Apr 18, 2024 | The p.Q227E variant (also known as c.679C>G), located in coding exon 6 of the BRIP1 gene, results from a C to G substitution at nucleotide position 679. The glutamine at codon 227 is replaced by glutamic acid, an amino acid with highly similar properties. This alteration has been identified in breast cancer patients as well as healthy controls across several studies (Easton DF et al. J. Med. Genet. 2016 05;53:298-309; Moyer CL et al. Cancer Res. 2020 Feb;80:857-867; Akcay IM et al. Int J Cancer, 2021 Jan;148:285-295; Dorling et al. N Engl J Med. 2021 02;384:428-439). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |

Familial cancer of breast;C1836860:Fanconi anemia complementation group J Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Jan 28, 2024 | This sequence change replaces glutamine, which is neutral and polar, with glutamic acid, which is acidic and polar, at codon 227 of the BRIP1 protein (p.Gln227Glu). This variant is present in population databases (rs45459799, gnomAD 0.008%). This missense change has been observed in individual(s) with breast cancer (PMID: 26921362, 31822495). ClinVar contains an entry for this variant (Variation ID: 216800). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt BRIP1 protein function with a negative predictive value of 80%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. - |
