The p.I146M variant (also known as c.438A>G), located in coding exon 4 of the BRIP1 gene, results from an A to G substitution at nucleotide position 438. The isoleucine at codon 146 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is not conserved however, methionine is a reference amino acid in several species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
