The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. - |

The proline at codon 47 is replaced by threonine, an amino acid with highly similar properties. This alteration was identified in 1/10030 consecutive patients referred for evaluation by an NGS hereditary cancer panel (Susswein LR et al. Genet. Med. 2016 08;18:823-32). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |

This variant has not, to our knowledge, been published in the literature as a pathogenic or benign germline variant. BRIP1 Pro47Thr was not observed in large population cohorts (Lek 2016). This variant is located in the helicase domain I (Cantor 2001). In silico analysis, which includes protein predictors and evolutionary conservation, supports a deleterious effect. Based on currently available evidence, it is unclear whether BRIP1 Pro47Thr is a pathogenic or benign variant. We consider it to be a variant of uncertain significance. - |
