The p.M29I variant (also known as c.87G>T), located in coding exon 1 of the BRIP1 gene, results from a G to T substitution at nucleotide position 87. The methionine at codon 29 is replaced by isoleucine, an amino acid with highly similar properties. This variant was identified in prostate and pancreatic cancer patients, as well as, controls of Japanese ancestry (Momozawa Y et al. J Natl Cancer Inst, 2020 Apr;112:369-376; Mizukami K et al. EBioMedicine, 2020 Oct;60:103033). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
