They are responsible for the generation and propagation of action potentials in neurons and muscle. This gene encodes one member of the sodium channel alpha subunit gene family. This residue falls in the repeat IV of the protein (UniProt) which plays a critical role in sodium voltage-gated channel alpha subunit 4's function. This is a previously reported variant (ClinVar 577938) that has been observed in individuals affected by myotonic disorder (PMID: 32670189). This variant is present in 22 of 402194 alleles (0.0055%) in the gnomAD population dataset. Multiple bioinformatic tools predict that this isoleucine to methionine amino acid change would be damaging, and the Ile1462 residue at this position is highly conserved across the vertebrate species examined. Studies examining the functional consequence of this variant have not been published, to our knowledge. At this time, there is insufficient evidence to determine if this variant is pathogenic or benign. Therefore, we consider this a variant of uncertain significance. ACMG Criteria: PM2, PP3 - |
