Variant summary: SCN4A c.952T>C (p.Trp318Arg) results in a non-conservative amino acid change located in the ion transport domain (IPR005821) of the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 0.00062 in 249302 control chromosomes (gnomAD). To our knowledge, no occurrence of c.952T>C in individuals affected with Acetazolamide-Responsive Myotonia and no experimental evidence demonstrating its impact on protein function have been reported. Eight clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 and classified the variant as benign/likely benign (n=6) and VUS (n=2) Based on the evidence outlined above, the variant was classified as uncertain significance. -
