The p.I519M variant (also known as c.1557C>G), located in coding exon 5 of the AXIN2 gene, results from a C to G substitution at nucleotide position 1557. The isoleucine at codon 519 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.

This sequence change replaces isoleucine, which is neutral and non-polar, with methionine, which is neutral and non-polar, at codon 519 of the AXIN2 protein (p.Ile519Met). This variant is present in population databases (rs374444786, gnomAD 0.004%). This variant has not been reported in the literature in individuals affected with AXIN2-related conditions. ClinVar contains an entry for this variant (Variation ID: 127936).
