This variant is present in population databases (rs774353403, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with AXIN2-related conditions. ClinVar contains an entry for this variant (Variation ID: 408818). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Not Available"; PolyPhen-2: "Benign"; Align-GVGD: "Not Available". The asparagine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. The p.S329N variant (also known as c.986G>A), located in coding exon 3 of the AXIN2 gene, results from a G to A substitution at nucleotide position 986. The serine at codon 329 is replaced by asparagine, an amino acid with highly similar properties. This alteration was observed in the homozygous state in a female child born to first-cousin parents of Turkish origin with a diagnosis of lymphomatoid granulomatosis; this individual was also found to have a homozygous FCHO1 pathogenic variant (Calzoni E et al. J Allergy Clin Immunol, 2019 06;143:2317-2321.e12). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
