Experimental studies have shown that this missense change affects KCNJ2 function (PMID: 17568571). For these reasons, this variant has been classified as Pathogenic. This variant disrupts the p.Asp78 amino acid residue in KCNJ2. Other variant(s) that disrupt this residue have been observed in individuals with KCNJ2-related conditions (PMID: 16217063), which suggests that this may be a clinically significant amino acid residue. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. ClinVar contains an entry for this variant (Variation ID: 67566). This missense change has been observed in individual(s) with Andersen-Tawil syndrome (PMID: 16419128, 17568571). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces aspartic acid, which is acidic and polar, with tyrosine, which is neutral and polar, at codon 78 of the KCNJ2 protein (p.Asp78Tyr). -
