Genes affected
KCNJ2 (HGNC:6263): (potassium inwardly rectifying channel subfamily J member 2) Potassium channels are present in most mammalian cells, where they participate in a wide range of physiologic responses. The protein encoded by this gene is an integral membrane protein and inward-rectifier type potassium channel. The encoded protein, which has a greater tendency to allow potassium to flow into a cell rather than out of a cell, probably participates in establishing action potential waveform and excitability of neuronal and muscle tissues. Mutations in this gene have been associated with Andersen syndrome, which is characterized by periodic paralysis, cardiac arrhythmias, and dysmorphic features. [provided by RefSeq, Jul 2008]

Andersen Tawil syndrome;C1865018:Short QT syndrome type 3 Pathogenic:1
Pathogenic, criteria provided, single submitterclinical testingInvitaeJan 31, 2024This sequence change creates a premature translational stop signal (p.Arg228*) in the KCNJ2 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 200 amino acid(s) of the KCNJ2 protein. This variant is present in population databases (no rsID available, gnomAD 0.003%). This premature translational stop signal has been observed in individual(s) with clinical features of KCNJ2-related conditions (Invitae). ClinVar contains an entry for this variant (Variation ID: 403974). This variant disrupts a region of the KCNJ2 protein in which other variant(s) (p.Ser369*) have been determined to be pathogenic (PMID: 21493816). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic. -
