Uncertain significance, criteria provided, single submitter | clinical testing | Ambry Genetics | Mar 07, 2017 | The p.I251M variant (also known as c.753C>G), located in coding exon 6 of the TP53 gene, results from a C to G substitution at nucleotide position 753. The isoleucine at codon 251 is replaced by methionine, an amino acid with highly similar properties. This alteration has been reported in a 50y/o patient diagnosed with anaplastic astrocytoma. The father of this patient was diagnosed with a glioblastoma at age 66y and a paternal cousin was diagnosed with a glioma at age 27y (Li YJ et al. Int J Cancer. 1995 Dec 20;64(6):383-7). This alteration has been reported as a germline mutation once, but not as a somatic mutation by the IARC TP53 database (Petitjean A et al. IARC TP53 database [version R17, November 2013]. Hum Mutat. 2007 Jun;28:622-9). This variant is in the DNA binding domain of the TP53 protein and is reported to have partial loss of transactivation capacity and dominant negative effect in yeast based assays (IARC TP53 database; Kato S et al. Proc Natl Acad Sci USA. 2003 Jul;100:8424-9; Malcikova J et al. Biol Chem. 2010 Feb-Mar;391(2-3):197-205; Monti P et al. Mol Cancer Res. 2011 Mar;9:271-9). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
