This premature translational stop signal has been observed in individual(s) with Li-Fraumeni syndrome (PMID: 9569035). ClinVar contains an entry for this variant (Variation ID: 12377). For these reasons, this variant has been classified as Pathogenic.

The c.532delC pathogenic mutation, located in coding exon 4 of the TP53 gene, results from a deletion of one nucleotide at nucleotide position 532, causing a translational frameshift with a predicted alternate stop codon (p.H178Tfs*69). This mutation has been reported in a kindred satisfying classic criteria for Li-Fraumeni syndrome (Sedlacek Z, Br. J. Cancer 1998 Apr; 77(7):1034-9). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
