Li-Fraumeni syndrome 1 Pathogenic:1
This variant is considered likely pathogenic. This variant is expected to disrupt protein structure [Myriad internal data]. Functional studies indicate this variant impacts protein function [PMID: 29979965]. -
Li-Fraumeni syndrome Pathogenic:1
For these reasons, this variant has been classified as Pathogenic. This sequence change replaces cysteine with arginine at codon 135 of the TP53 protein (p.Cys135Arg). The cysteine residue is highly conserved and there is a large physicochemical difference between cysteine and arginine. This variant is not present in population databases (ExAC no frequency). This variant has been observed in individual(s) with clinical features of Li Fraumeni disease (PMID: 31050713, Invitae). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 376560). This variant has been reported to affect TP53 protein function (PMID: 12826609, 30224644). -
