Mutations in this gene are associated with a variety of human cancers, including hereditary cancers such as Li-Fraumeni syndrome. Alternative splicing of this gene and the use of alternate promoters result in multiple transcript variants and isoforms. Additional isoforms have also been shown to result from the use of alternate translation initiation codons from identical transcript variants (PMIDs: 12032546, 20937277). The variant was observed in the following populations: Other in 1 of 6414 chromosomes (freq: 0.0002), Latino in 4 of 34336 chromosomes (freq: 0.0001), European in 2 of 125870 chromosomes (freq: 0.00002), East Asian in 100 of 18800 chromosomes (freq: 0.005), and South Asian in 1 of 30676 chromosomes (freq: 0.00003), while the variant was not observed in the African, Ashkenazi Jewish, and Finnish, populations.
