The c.3097G>A (p.E1033K) alteration is located in exon 20 (coding exon 19) of the MYOM1 gene. This alteration results from a G to A substitution at nucleotide position 3097, causing the glutamic acid (E) at amino acid position 1033 to be replaced by a lysine (K). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 1033 of the MYOM1 protein (p.Glu1033Lys). This variant is present in population databases (rs377512048, gnomAD 0.06%), and has an allele count higher than expected for a pathogenic variant. This variant has not been reported in the literature in individuals affected with MYOM1-related conditions. ClinVar contains an entry for this variant (Variation ID: 454444).
