Experimental studies have shown that this missense change results in this variant fails to bind DNA and may result in insoluable aggregates of TFIF1 protein compared to wildtype (PMID: 16962354). This variant has been reported to be de novo in individuals affected with holoprosencephaly (PMID: 11810641). This sequence change replaces arginine with cysteine at codon 90 of the TGIF1 protein (p.Arg90Cys). The arginine residue is highly conserved and there is a large physicochemical difference between arginine and cysteine. This variant is not present in population databases (ExAC no frequency). In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic. -
