The c.7082A>C (p.E2361A) alteration is located in exon 41 (coding exon 41) of the EPG5 gene. This alteration results from a A to C substitution at nucleotide position 7082, causing the glutamic acid (E) at amino acid position 2361 to be replaced by an alanine (A). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
Vici syndrome Uncertain:1
This sequence change replaces glutamic acid with alanine at codon 2361 of the EPG5 protein (p.Glu2361Ala). The glutamic acid residue is moderately conserved and there is a moderate physicochemical difference between glutamic acid and alanine. This variant is present in population databases (rs374750937, ExAC 0.03%). This variant has not been reported in the literature in individuals affected with EPG5-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
