The p.I482V variant (also known as c.1444A>G), located in coding exon 10 of the SMAD4 gene, results from an A to G substitution at nucleotide position 1444. The isoleucine at codon 482 is replaced by valine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
Juvenile polyposis/hereditary hemorrhagic telangiectasia syndrome Uncertain:1
This missense variant replaces isoleucine with valine at codon 482 of the SMAD4 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373).
